So, I have figured out how to represent the 100 meter boxes. I just added tick marks in the Data Frame>Grids>Properties and just made them subdivisions of 10, made the tick marks point inward, as opposed to the outside of the layout, and expanded the tick marks to go across the whole map. Then I just added "a ladder of interior labels" to help reference where you are inside the 1,000 meter box. It does the job.

Hopefully you found the NGA geodatabase that covers 133 cities with a fine MGRS grid.
The geodatabase contains MGRS grids at 500m and 100m.

I am attempting to create a 1000m grid that will match the grid lines on a TLM50 NGA raster map product. The data frame coordinate system is GCS_WGS_1984, datum is D_WGS_1984. When I create a measured grid with 1000m intervals, the grid lines produced do not match the map background grid lines. ???
Try projecting the data frame to the same UTM zone as the TLM.
